I'll leave it open ended and open to interpretation.
The one that I can think of is week 5 against the Browns in 2008. What everyone remembers is how dominant we looked until Burress went down, I think we were 10-1 (eventually went to 11-1 and then 12-4), but no one remembers that one loss. The Browns in 2008 were 4-12. Not only did the Giants lose they got their asses handed to them, I don't think the Browns punted once in that game. I still can't figure it out.
If there wasn't a fix in - then that was the most incompetent string of calls made in a long time, and that includes the Packers playoff game.
I honestly don't think I've seen that defense again since that game.
I thought the season was going to fall apart after that game. But that team just kept hammering opponents. Its' too bad attrition and plain old stupidity derailed that team. Back to back would have been awesome.
I guess this is the obvious answer that I didn't even think of. Was originally going to limit it to regular season games. But yeah, I guess this is #1.

That loss forced the Giants to have to beat both the Jets and Dallas in the final 2 weeks.

I was thinking of these exact games plus there was a game where we played Tennessee and we had a 21-0 lead. I think the game turned when Kiwanuka let go of Vince Young because he might have been afraid of roughing the passer.
and of course that dreaded 2010 game before Christmas.
I was too young in 2002 to be too destroyed by it but I think that might have been the most painful game I experienced as a fan, at least NFL game.

It's scary to think but we were a Lawrence Tynes miss in 2007 from having that game X 100.
Meh. This shouldn't be on anyone's list who is a Giants fan. The way Manninghams fumble perfectly rolled on the ground in bounds should've been a clear ominous sign that game had Philly comeback written all over it.
I don't even blame Matt Dodge for that game. Coughlin should've been in his defense's face at the end.
We played them in preseason that year and absolutely and utterly embarrassed them. It was like the Oklahoma Sooners playing Western Tulsa Community college.
We played them at home and we took them WAY lightly, and they were out for revenge big time. We never came anywhere near matching their intensity that game. That was like a Super Bowl for them.
Most of these are games we lost, but I'll choose a game we actually won, the eagles finale in 2002. We had 7 fumbles and several bs penalties that negated TDs and big gains. I remember even the announcers were getting skeptical after the bomb by Collins was called back. It seemed like fate was against us that day. In a game that we should have won by 40 pts we won 10-7...in OT.

I remember that game. I was strangely more upset over that game than the playoff game. I remember going to a family party that night distraught. I didn't think people would remember it. I think a similar game was against the Chargers, I think in 2009.

Kind of embarrassing to admit but when Shawn Merriman did his "lights out" dance on the final play I threw my keys at the TV and broke it.

The Giants began the season losing three straight. I still remember Frank Gifford, in his comments just prior to game three (he was doing the game for CBS) saying that it was almost impossible for a team starting 0-3 to make the playoffs.
The Giants lost that third game to a Saints team that would finish the season with the worst record in the NFL... 2-11-1.
But then the Giants, led by Fran Tarkenton and Ron Johnson, reeled off 6 straight wins heading into their very first Monday Night game against the Eagles.
The Eagles came into the Monday Night game with a 1-7-1 record and ended the season with the second-worst record in the NFL.
The Giants had won 6 straight. The Eagles sucked! I was already counting it as a win. But of course the Giants lost.
The Giants then won their next 3 games and entered the final week of the season with their fate in their own hands.
Beat the Rams and they win the East. Lose and they're out of the playoffs. (In those days just four teams made the playoffs in each conference...the three division winners and one WC.)
I started rooting for the Giants in '65. It was going to be my first playoffs.
All they had to do was beat the Rams.
Final Score: Rams 31 Giants 3.

Never gave the Giants a shot vs the 13 0 Broncos.
Because the Giants failed the two point conversion attempt, this allowed Tennessee to only need a touchdown and a two-point conversion of their own to tie the score at 29-29.

The Seahawks in 86 were the bet team in the NFL not to make the playoffs. They weren't scrubs. They were the only team to beat both Super Bowl participants in the 1986 season.
I think up until New England got blown out by Kansas City in 2017, the Giants were the only team to lose Week 1 at home in the kickoff game.
That was the worst for me; especially as a season ticket holder sitting in the stands. I still remember Phillips going for the pick and whiffing on the first Eagle touchdown and getting an uneasy feeling in my gut. Then they got the onside kick and I knew we were in trouble.
I always had the most confidence in the Giants against good teams back then - and I always felt like we were going to lose games we should have won.
It often seemed to go that way.

I remember that, the only drive Minnesota did anything was on that last drive.
The shootout in New Orleans where we actually took the lead with 7 minuteds left only to blow it at the end because of a face mask penalty on a punt.
The Jets game where we had the lead late only to let the Jets march down the field in the final 4 minutes for the tying TD and lose in OT
The Patriots game where Beckham could not hold on for a TD that would have forced the Pats to have to score a TD in the final 2 minutes instead of a FG to beat us.
