But I will give it a try. I'm including only pure comedies (so romantic comedies (a favorite genre of mine) are left out). In addition, I considered historical and influential context in the equation.
Duck Soup
Monty Python and the Holy Grail
Bananas
Airplane
There were a lot of strong contenders (some of which have already been mentioned in this thread) that could easily replace any one of these. In other words I could probably come up with another four equally deserving of the honor in my mind. Removing romantic comedies (Annie Hall, It Happened One Night, The Apartment, His Girl Friday/The Front Page, When Harry Met Sally) and dramedies (Dr. Strangelove, Crimes & Misdemeanors) from the mix helps, but not nearly enough.
I went with Airplane over The Naked Gun, but I think Naked Gun is funnier; it's just that Airplane came first. It's also why I chose Bananas over Sleeper and Love & Death (and there absolutely needed to be a Woody Allen movie on Mount Rushmore!). I know Take the Money and Run came before Bananas, but Woody hadn't yet come into his own as a writer/director until Bananas (he of course would continue to improve as a filmmaker).

Was waiting for Animal House. Unless you're too young it was instantly a cultural phenomenon when it came out. And FFS BBI catch phrase "Germans". lol.

You know, Pootie Tang was born in a small town outside of Gary, Indiana. And that town is called Chicago.
